They are very hygienic. Most cups (including the Lunette) are made from high quality medical grade silicone, which naturally prevents any bacteria from growing. A quick rinse after emptying and reinserting is fine, and washing with a non irritating soap once a day is perfect. Sterilize between cycles for even more confirmation (though just washing is fine). I've had my Lunette since August of 2012 (so nearly 4 years), and I've had no complications / infections even with very heavy and repeated usage. It really is great, I love it.

Once the black sets, it is in place. It comes off easily with coconut oil or makeup remover. I like it because it doesn't annoy my eyes that tend to water when tightlining. This is the best waterproof eyeliner for the drugstore price tag.

The lighter colors don't seem to be as pigmented, but the darker colors can be very BAM! on the lips and will stain once the top layer has worn off. I recommend Honey as a neutral shade and Crush and Smitten as darker more staining shades.
